Hi Sonny,
I suspect the key challenge will be mounting your cells in the OpenSPIM
geometry. But there are light sheet microscopes built around a
horizontal sample mount, including diSPIM, lattice light sheet, and
Leica's confocal add-on.
For the diSPIM (http://dispim.org/) it is possible to use a NA 1.0 or
1.1 objective if the usual 0.8 NA isn't enough. It's also possible to
incorporate Bessel beam, etc. I'm happy to answer further questions but
we should move off this mailing list.
